The impact of carbon source in Candida albicans virulence: participation of RLM1 in pathogen host interaction
Dissertação de mestrado em Genética MolecularThe survival of all microbes depends upon their ability to respond to environmental
challenges. To establish infection, pathogens such as Candida albicans must support effective
stress responses to counter host defenses while adapting to dynamic changes in nutrient status
within host niches. Studies of C. albicans stress adaptation have generally been performed on
glucose-grown cells, leaving the effects of alternative carbon sources upon stress resistance largely
unexplored as well alterations in its virulence.
In a previous work both copies of the RLM1 gene of C. albicans were deleted and phenotypic
analysis of Δrlm1/Δrlm1 mutants showed typical cell wall weakening phenotypes, such as
hypersensitivity to Congo Red, Calcofluor White and caspofungin, showing its involvement in the
cell wall remodelling. To understand the role of RLM1 under the influence of different carbon
source, phenotypic characterization against cell wall damaging stress agents, modulation of
virulence factors as well as their involvement in host-pathogen interaction was preformed in the
present work.
Candida albicans Δrlm1/Δrlm1 mutant displayed phenotypes associated to cell wall
deficiency such as, hypersensitivity to Congo red, caspofungin in glucose- and lactate- grown cells.
However the RLM1 mutants were slight more resistant to Congo Red when grown in lactate. On the
other hand lactate-grown cells were not able to growth al all in the presence of SDS and presented
sensitivity to caffeine, in comparison with glucose-grown cells. The increased transcription of genes
already reported to be involved in cell adhesion correlated well with adhesion and biofilm assays, in
which RLM1 mutant presented greater biofilm formation than WT in both growth condition.
However cell adapted to lactate adhered more and biofilm formation was more pronounced. The
host-pathogen interaction was accessed by co-incubation with murine macrophages-like cell line
(J774). In general, lactate-grown cells were less efficiently killed in comparison to glucose-grown
cells. However the mutant presented distinct behaviors, they were more resistant when adapted to
glucose than to lactate. The TNF-α and IL-10 were lower in response to Δrlm1/Δrlm1 mutant and
the cellular toxicity, measured as extracellular lactate dehydrogenase activity, was significantly
lower in comparison with the WT and complemented strains in glucose-grown cells. The effect
observed before was reverted when C. albicans cells were grown on lactate.
In conclusion, C. albicans cells adapted to different carbon sources behave differently,
particularly in the interaction with macrophages, in which the RLM1 mutation plays a decisive
effect since it affects the cell wall integrity.Os microrganismos para sobreviver dependem da capacidade de responder aos desafios
apresentados pelo ambiente onde estão inseridos. O sucesso da infecção por parte dos agentes
patogénicos, tais como Candida albicans, assenta na capacidade de responder eficazmente a
stresses causados pelas defesas do hospedeiro e à adaptação a alterações constantes dos
nutrientes no hospedeiro. Alguns estudos indicam que o crescimento em fontes de carbono
alternativas tem influência na resistência ao stress e alteram a virulência de C. albicans.
Em trabalhos anteriores, o gene RLM1 de C. albicans foi deletado e a análise fenotípica dos
mutantes Δrlm1/Δrlm1 revelou que estes apresentam hipersensibilidade ao Congo Red,
Calcofluor white e Caspofungina, fenótipos típicos de fragilidade na parede celular. De forma a
perceber o papel de RLM1 em células adaptadas a diferentes fontes de carbono, foi feita uma
caracterização fenotípica usando agentes que perturbam a parede celular, avaliada a modulação
de alguns fatores de virulência, bem como a interação hospedeiro-patogéno.
Os mutantes Δrlm1/Δrlm1 de Candida albicans crescidos em glucose e láctico exibiram
fenótipos associados a deficiências na parede celular, tais como, hipersensibilidade ao Congo Red
e Caspofungina. No entanto, estas revelaram ser ligeiramente mais resistentes ao Congo Red
quando crescidas em láctico. Por outro lado, as células crescidas em ácido láctico apresentaram
maior dificuldade em crescer na presença de cafeína, não tendo sido detectado crescimento em
SDS, contrariamente às células crescidas em glucose que não apresentaram sensibilidade a estes
stresses. O aumento da expressão de genes envolvidos na adesão célular correlacionou-se
diretamente com os ensaios de adesão e biofilme, em que os mutantes Δrlm1/Δrlm1 apresentam
uma maior formação de biofilme em comparação com a estirpe selvagem em ambas condições de
crescimento. Contudo, células crescidas em ácido láctico aderiram mais e a formação de biofilme
foi mais evidente. A interação Candida-hospedeiro foi avaliada através da co-incubação com a linha
celular de macrófagos J774. Em geral, as células crescidas em ácido láctico foram fagocitadas de
forma menos eficiente em comparação com células crescidas em glucose. As estirpes mutante
Δrlm1/Δrlm1 apresentaram comportamentos distintos quando crescidas em diferentes fontes de
carbono; em glucose os níveis de produção de TNF-α e IL-10 foram mais baixos e a toxicidade
celular, avaliada pela atividade da lactato desidrogenase, foi significativamente mais baixa, em
comparação com as estirpes selvagem e complementadas. O efeito anteriormente observado foi
totalmente revertido quando o mutante foi crescido em meio com ácido láctico.
Em conclusão, células de C. albicans a diferentes fontes de carbono apresentam
comportamento diferentes, nomeadamente na interação com macrófagos, onde o gene RLM1
parece ter um papel decisivo pois afecta a integridade da parede celular
Aspetos de modelos Einstein-Maxwell-scalar: solitões, dualidade e escalarisação
In the recent years, the Einstein-Maxwell-scalar model has proven to be a very fruitful and interesting model. Due to the simple addition of a non-minimal coupling f(φ) between the scalar field φ and the electromagnetic field terms, we find various novel and unusual properties that have led to interesting results in the fields of general relativity (at least as a toy model) in the astrophysics of compact objects. This thesis will address some of these properties. After a brief introduction of the topics to be covered, we present the various results that we obtained regarding the non-existence of solitons in certain conditions for this model. These results paved the way for us to obtain a new soliton solution for this kind of model, which is also presented here. Next, we discuss the duality symmetries of the Einstein-Maxwell-scalar model and take advantage of them to create a solution generating technique which we then apply to wellknown solutions of the model. Lastly, we cover the much discussed concept of spontaneous scalarisation, but this time in higher dimensions for a generalised model. We end with some conclusions and remarks.Nos anos recentes, o modelo de Einstein-Maxwell-scalar provou ser
um modelo bastante rico e interessante. Devido à simples adição
de um acoplamento não minimal f(φ) entre o campo escalar φ e o
campo electromagnético, encontramos várias propriedades peculiares
que levaram a resultados interessantes nos campos da relatividade
geral e (pelo menos como um modelo teste) da astrofísica de objetos
compactos. São algumas destas propriedades que vamos discutir nesta
tese.
Após uma pequena introdução dos tópicos que vão ser abordados,
apresentamos vários resultados que obtemos sobre a inexistência de
solitões para certas condições neste modelo. Estes resultados abriram
o caminho para obtermos uma nova solução solitónica para este tipo
de modelo, que também é apresentada aqui. A seguir, discutimos as
simetrias de dualidade do modelo Einstein-Maxwell-scalar e tiramos
partido destas para obter uma técnica de geração de soluções que
depois aplicamos a soluções conhecidas deste modelo. Por m, abordamos o muito discutido conceito de escalarisação espontânea, mas
desta vez em espaços de alta dimensão para um modelo generalizado.

Reaction-diffusion systems derived from kinetic models for Multiple Sclerosis
We present a mathematical study for the development of Multiple Sclerosis in
which a spatio-temporal kinetic model describes, at mesoscopic level, the
dynamics of a high number of interacting agents. We consider both interactions
among different populations of human cells and motion of immune cells,
stimulated by cytokines. Moreover, we reproduce the consumption of myelin
sheath due to anomalously activated lymphocytes and its restoration by
oligodendrocytes. Successively, we fix a small time parameter and assume that
the considered processes occur at different scales. This allows to perform a
formal limit, obtaining macroscopic reaction-diffusion equations for the number
densities with a chemotaxis term. A natural step is then to study the system,
inquiring about the formation of spatial patterns through a Turing instability
analysis of the problem and basing the discussion on microscopic parameters of
the model. In particular, we get spatial patterns oscillating in time that may
reproduce brain lesions characteristic of different phases of the pathology
Simulated visually-guided paw placement during quadruped locomotion
Autonomous adaptive locomotion over irregular terrain
is one important topic in robotics research. In this article, we
focus on the development of a quadruped locomotion controller
able to generate locomotion and reaching visually acquired
markers. The developed controller is modeled as discrete, sensory
driven corrections of a basic rhythmic motor pattern for
locomotion according to visual information and proprioceptive
data, that enables the robot to reach markers and only slightly
perturb the locomotion movement. This task involves close-loop
control and we will thus particularly focus on the essential issue of
modeling the interaction between the central nervous system and
the peripheral information in the locomotion context. This issue
is crucial for autonomous and adaptive control, and has received
little attention so far. Trajectories are online modulated according
to these feedback pathways thus achieving paw placement. This
modeling is based on the concept of dynamical systems whose
intrinsic robustness against perturbations allows for an easy
integration of sensory-motor feedback and thus for closed-loop
control.
The system is demonstrated on a simulated quadruped robot
which online acquires the visual markers and achieves paw
placement while locomotes
Too old for technology?:age stereotypes and technology use by older adults
Information and communication technology holds great promise in supporting the growing population of older people. However, older adults are less likely to use technology compared to the general population, limiting their ability to live independently in an increasingly digital world. Besides being commonly stereotyped as less technologically competent than younger age groups, older adults often mention their age or being “too old” as reasons for not using technology. Surprisingly, the potential influence of stereotypical perceptions about older people and the ageing process on older adults’ use behaviours remains largely understudied. This work aimed to fill this gap by investigating whether and how age stereotypes relate to technology use in late adulthood. Findings from six studies suggest that age stereotypes determine technology use among older age groups through distinct, yet complementary processes. Interventions and policies promoting positive intergenerational contact and positive views on age and ageing should therefore contribute to the digital inclusion of older adults

Older adults are less likely to use information and communication technologies, limiting their ability to live independently in an increasingly digital world. Despite being stereotyped as lacking technological competence, the influence of age stereotypes on older adults’ use behaviours remains largely understudied. This work aimed to investigate whether and how age stereotypes determine technology use in late adulthood. According to stereotype threat theory, the fear of confirming negative stereotypes about their age group may lead older adults to avoid using technology. Across three studies based on longitudinal and cross-sectional designs, higher levels of stereotype threat were associated with lower rates of technology use. In line with the
technology acceptance model, this relationship was mediated by anxiety, perceived ease of use, perceived usefulness, and behavioural intention. In turn, as implied by stereotype embodiment theory, the internalization of age stereotypes endorsed earlier in life into self-perceptions of ageing later in life may deter older adults from using technology. Across three studies based on longitudinal data from the Health and Retirement Study and the German Ageing Survey, more positive self-perceptions of ageing were associated with higher levels of technology use.
Cognitive functioning mediated this relationship. By confirming the assumptions of both theories, these findings suggest that age stereotypes influence technology use in late adulthood through complementary processes. Future studies should further explore these relationships by integrating both theoretical approaches. Interventions and policies promoting positive intergenerational contact and positive views on age and ageing should thus contribute to the digital inclusion of older adults.As pessoas mais velhas têm uma menor probabilidade de usar tecnologias de informação e comunicação, impedindo que vivam de forma independente num mundo cada vez mais digital.
Apesar de serem estereotipadas como tendo menos competências tecnológicas, a influência destes estereótipos nos seus padrões de utilização permanece pouco estudada. Este trabalho pretendeu investigar se e como os estereótipos etários determinam os comportamentos de utilização de tecnologias das pessoas mais velhas. Segundo a teoria da ameaça do estereótipo, o receio de confirmar estereótipos negativos pode fazer com que os mais velhos evitem usar tecnologias. Três estudos longitudinais e correlacionais verificaram uma relação entre maiores níveis de ameaça do estereótipo e menores taxas de utilização de tecnologias, sendo esta mediada pela ansiedade, facilidade percebida, utilidade percebida e intenção comportamental de acordo com o modelo de aceitação de tecnologias. Por sua vez, segundo a teoria da incorporação do estereótipo, a internalização de estereótipos etários em autoperceções de envelhecimento pode demover os mais velhos de usar tecnologias. Três estudos longitudinais verificaram uma relação entre autoperceções de envelhecimento mais positivas e maiores níveis de utilização de tecnologias, sendo esta mediada pelo funcionamento cognitivo. Ao confirmar os pressupostos das duas teorias, estes resultados sugerem que os estereótipos etários influenciam os comportamentos de utilização de tecnologias das pessoas mais velhas através de processos complementares. Estudos futuros deverão explorar estas relações integrando ambas as teorias. Intervenções que promovam contactos intergeracionais positivos e visões positivas sobre a idade e o envelhecimento poderão contribuir para a inclusão digital deste grupo etário

Dissertação para a obtenção do Mestrado em Direito Internacional e EuropeuSpace is increasingly a potential stage for conflict, and at the same time humanity
grows more dependent from space assets. Moreover, the current legal framework
does not place any constraint on the use or testing of conventional weaponry in
outer space.
The question of the regulation of conventional military activities in space was
discussed mainly under the concept of Prevention of an Arms Race in Outer Space
(PAROS) since the 1980s. The two main proposals for a legal framework for this
issue are the draft Treaty on the Prevention of the Placement of Weapons in Outer
Space and of the Threat or Use of Force against Outer Space Objects (PPWT) and
the draft International Code of Conduct for Outer Space Activities (Code), which
demonstrate fundamental differences with regard to key definitions, the
interpretation of PAROS and the general approach to the regulation of
conventional military activities in space.
The PPWT (proposed by Russia and China) represents the classic version of
PAROS, that is, a hard law approach to implement an arms control regime
negotiated in a multilateral forum. It failed because multilateral forums are blocked
in the post-Cold War world, as well as due to its highly criticized definition of
space weapon and the absence of verification mechanisms which deprived it of the
potential benefits of the hard law approach.
The Code (proposed by the EU) represents a first update in the approach to the
regulation of conventional military activities in outer space, as well as in the
interpretation of the PAROS issue. It proposes a soft law instrument negotiated
bilaterally to implement norms focused on preventing behaviour that causes space
debris. The Code failed because it hinged too much on soft law to avoid providing
key definitions, and on the common interests of States in preventing the “tragedy
of the commons”.
Deterrence is considered by the US to be the best way to guarantee its national
security, therefore it is reluctant to change the status quo. However, this strategy

is not without its flaws and since both the PPWT and the Code have stagnated, a
political vacuum has emerged that can be filled by the US as the lead norm builder
for security in space. In this sense, although the Artemis Accords are not intended
to be a response to PAROS, they can still be a glimpse of an alternative approach
to formulating norms for conventional military activities in space.A humanidade depende cada vez mais do espaço e ao mesmo tempo a possibilidade
de conflito no mesmo também aumenta; além disso, o atual quadro jurídico não
impõe restrições ao uso ou teste de armamento convencional no espaço.
A questão da regulamentação das atividades militares convencionais no espaço foi
discutida principalmente sob o conceito da Prevenção da Corrida ao Armamento
no Espaço (PAROS) desde a década de 1980. As duas principais propostas de
regulamentação são o Tratado para a Prevenção da Colocação de Armas no Espaço
e da Ameaça ou Uso da Força contra Objetos Espaciais (PPWT) e o Código
Internacional de Conduta para Atividades no Espaço (Código de Conduta), que
demonstram diferenças fundamentais no que diz respeito a definições-chave, à
interpretação da PAROS e abordagem geral à regulação das atividades militares
convencionais no espaço.
O PPWT (proposto pela Rússia e China) representa a versão clássica da PAROS,
ou seja, uma abordagem de hard law para implementar um regime de controlo de
armas negociado em fóruns multilaterais. O PPWT falhou porque os fóruns
multilaterais estão bloqueados no mundo pós-Guerra Fria, bem como devido à sua
definição de arma espacial que foi altamente criticada e a ausência de mecanismos
de verificação que o privaram dos benefícios de ser uma proposta hard law.
O Código de Conduta (proposto pela UE) representa uma primeira atualização na
abordagem da regulamentação das atividades militares convencionais no espaço,
bem como na interpretação da questão PAROS. Propõe um instrumento soft law
negociado bilateralmente com vista à implementação de normas que previnam a
criação de detritos espaciais. O Código fracassou por depender excessivamente da
soft law de maneira a evitar o fornecimento de definições-chave e nos interesses
comuns dos Estados em prevenir a “tragédia dos comuns”.
A estratégia de dissuasão é considerada pelos EUA a melhor maneira de garantir
a sua segurança nacional, pelo que estão relutantes em alterar o status quo. No
entanto, não é isenta de falhas e visto que tanto o PPWT como o Código

estagnaram, surgiu um vácuo político que pode ser preenchido pelos EUA como o
principal criador de normas para a segurança no espaço. Nesse sentido, embora os
Acordos Artemis não tenham como objetivo ser uma resposta à PAROS, podem
ainda assim ser um vislumbre de uma abordagem alternativa à formulação de
normas para atividades militares convencionais no espaço
